I'm trying to read data() from a cell in a datatable which has a button inside it, but I'm getting en error.
This is my Datatable definition:

Why am I suggesting this: I have a pretty similar problem, where DataTables is not able to call the rowReorder event. I also create my table via AJAX / dynamically (which you seem to do aswell), so obviously my event listener (in my case for table.on('row-reorder')) was bound multiple times. After first removing the event listener and readding it, finally I got this to work.

I have a jquery autocomplete that once a value is selected it loads a datatable with checkbox from an ajax call. Then while submitting the form I need to access the datatable variable to iterate each row to get the selected ones, but the datatable variable appears as undefined.
I'm doing the same as in this example, only difference is the data is coming from an Ajax request.
Can you please help me understand why is that happening?

You may need to grab your DataTables object into a variable before using that:

var rows_selected = t_OmnitureCode.column(0).checkboxes.selected();
And, by the way, your method of populating DataTable with external ajax-call is suboptimal. There's an ajax option for that purpose where you can specify all the necessary parameters and get better integration with DataTables API and better performance (as you don't really need to destroy and create your DataTable upon each refresh).
You would simply need to trigger .ajax.reload() whenever you need to refresh your table data.
It's a matter of scope : You declare the variable table inside $(document).ready function.
You may want to put it outside in the global scope :

I have a couple of drop downs that are populated from SharePoint using SPServices. This part works great. But then, I have a button that on click loads data from SharePoint and uses the dropdown texts as filter to fetch the data that will populate a table using the DataTables plugin. This part works only once; if I click the button again, nothing happens.

I would guess that you are replacing the part of the page where the button lives. (you really need to format your code more neatly for SO... use JSFiddle.net and their TidyUp button).
If that is the case you need to use a delegated event handler:
$(document).on('click', '.myButton', function () {
run()
});
This listens at a static ancestor of the desired node, then runs the selector when the event occurs, then it applies the function to any matching elements that caused the event.
document is the fallback parent if you don't have a convenient ancestor. Do not use 'body' for delegated events as it has odd behaviour.

However - if I open Developer Tools in Chrome I get an error message saying sAjaxSource is null so i cannot then get the value from it - so oSettings is null and then I cannot get access to the sAjaxSource - anyone see anything wrong here?
you're basically reinitializing your dataTable in the first row of refreshTable. Try instead:
